Intratumoral Expression of Mature Human Neutrophil Peptide-1 Mediates Antitumor Immunity in Mice

Abstract: Purpose: Human neutrophil peptides (HNP1-3), small molecular antimicrobial peptides, are expressed within tumors and associated with tumor necrosis and inhibition of angiogenesis. Recent investigations have suggested that HNP1-3 are likely to be involved in the host immune responses to tumors. Experimental Design: We used recombinant pSec-HNP1, which expresses a secretable form of HNP1, to obtain expression of HNP1 in the tumor milieu in immunocompetent mice to explore the possible roles of HNP1 in tumor immun… Show more

“…27 As in our model, we used the 4T1 murine metastatic breast cancer cells, which can metastasize to the lung with high frequency in BALB/c mice as early as 2 weeks after inoculation. 25,28 Based on these findings, when the 4T1 tumor volume reached 2000 mm 3 in the current study, mice were statistically regarded as imminent deaths and killed. The metastatic lung nodules were counted under a dissecting microscope.…”

“…25 Nonnecrotic tumors were harvested from the mice in the various treatment groups described previously for cytokine evaluation. TNF-a, IL10 and IFN-g levels were measured in the supernatant of tumor homogenates by a cytokine-specific ELISA kit.…”

“…In our previous investigations, intratumorally expressed HNP1 exhibited multiple antitumor effects in vivo, and in particular, mediated host immune responses to tumors (18). Considering the impact of tumor burden on the potential efficacy of immunotherapy, in animal experiments, pHNP1 was administered when the tumor diameter reached 4 mm.…”

“…Previously, we observed that intracellularly expressed HNP1 increased plasma membrane permeability in human A549 lung carcinoma cells (17). We also noted that the intratumoral expression of HNP1 mediated specific angiogenesis inhibition in vivo (17,18). In addition, HNP1 can cause VEGF-dependent endothelial cell proliferation inhibition by forming a ternary complex with fibronectin and α 5 β 1 integrin (19).…”
