Intrinsic Diurnal Variations in Cardiac Metabolism and Contractile Function

Abstract: Abstract-Diurnal variation of cardiac function in vivo has been attributed primarily to changes in factors such as sympathetic activity. No study has investigated previously the intrinsic properties of the heart throughout the day. We therefore investigated diurnal variations in metabolic flux and contractile function of the isolated working rat heart and how this related to circadian expression of metabolic genes. Contractile performance, carbohydrate oxidation, and oxygen consumption were greatest in the mid… Show more

“…Blood pressure and heart rate were obtained throughout the study. PET was used to measure myocardial blood flow with 15 O-water, MVO 2 with 1-11 C-acetate, myocardial glucose extraction fraction and utilization with 1-11 C-glucose, and fatty acid extraction fraction, utilization, and oxidation with 1-11 C-palmitate. During the study, venous blood samples were obtained at predetermined intervals to measure plasma substrate (glucose, fatty acids, and lactate) and insulin levels as well as radiolabeled metabolites, which are required for compartmental modeling of the kinetics of the metabolic tracers.…”

“…13 Measurements of myocardial blood flow (MBF), MV O 2 , glucose uptake (MGUp) and utilization (MGU), and MFAUp, MFAU, and MFAO were obtained with PET with 15 O-water, 1-11 C-acetate, 1-11 C-glucose, and 1-11 C-palmitate, respectively, using validated techniques as reported previously. 14 -20 The interval between each radiopharmaceutical injection allowed for 5 half-lives of decay time of the preceding radiopharmaceutical.…”

“…This gene was selected because among all tested housekeeping genes (Gapdh, Hprt1, and Actb), it displayed the lowest variations between the various experimental conditions. To validate the method, we independently analyzed two genes (Pdk4 and Ucp3) using TaqMan quantitative RT-PCR, as described previously (51,52). Furthermore, Me2 and Pcx were also analyzed using TaqMan assays, as described previously (13).…”
