Invasion by cultured human follicular thyroid cancer correlates with increasedβ1 integrins and production of proteases

Abstract: A recognized model of tumor invasion requires cells to adhere to epithelial basement membrane and extracellular matrix components triggering release of proteases thus allowing cancer cells to invade the substrate. This adhesion is mediated by beta 1 integrins, a family of receptors to substrates such as collagen, laminin, and fibronectin. In order to study tumor invasion in follicular thyroid cancer (FTC), we used cell lines derived from a single patient's FTC primary tumor (FTC-133), neck lymph node metastase… Show more

“…Of the three cell lines, FTC236 showed a moderate invasive behavior and FTC238 a high invasive potential, whereas FTC133 did not (Demeure et al 1992, Hoelting et al 2001. The invasive behavior of these human thyroid cancer cell lines positively correlated with their CCL2 expression levels (Fig.…”

“…Aberrant expression of focal adhesion components, including integrins, FAK, and Rho GTPases, has been found in invasive thyroid cancers [Demeure et al, 1992;Owens et al, 1996;Schmitz et al, 2000], implying that focal adhesion might play an important role in the malignant phenotype of thyroid cancers.…”

“…FTC-238 is a cell line (ECACC, Salisbury, UK) derived from a lung metastasis of a follicular thyroid carcinoma from a 42-yearold man (Demeure et al 1992). BRA-2 is a thyroid cell line established in our laboratory in 1995 and derived from a lymph node metastasis of a papillary thyroid carcinoma from a 74-yearold woman.…”
