Inverse Correlation between Serum Testosterone and Leptin in Men1

Abstract: Besides its role in the regulation of energy balance, leptin seems to be involved in linking energy stores to the reproductive system. A gender-dependent difference exists in plasma leptin concentration and leptin messenger ribonucleic acid expression in rodents and humans. This difference does not seem to be explained simply by differences in the amount of body fat between genders. To elucidate the relationship of endogenous testosterone and leptin, we studied the serum leptin concentrations in 269 elderly no… Show more

“…In contrast, during the 12 months leptin levels in the placebo group increased by 15%. Similar results were obtained by Luukkaa et al in young healthy males who used 200 mg of testosterone intramuscularly once weekly for 12 months as a contraceptive [12]. After three months of testosterone withdrawal leptin levels returned to baseline, which may indicate direct inhibitory effects of testosterone on leptin production.…”

“…The baseline leptin concentrations of these men were not correlated with the BMI, but the correlation was considerably improved during testosterone replacement. Similarly, testosterone (200 mg testosterone enanthate, intramuscularly weekly for 12 months) decreased the plasma leptin level in 10 healthy men from 3.4 to 1.9 µg/l [6]. It returned to its initial value when testosterone was stopped.…”

“…Conversely, blocking testosterone secretion with gonadotropin-releasing hormone (GnRH) analog increased the plasma leptin concentrations of boys with precocious puberty, while resumption of puberty was associated with decreased leptin levels [12]. A similar increase was observed in 6 young men after gonadal suppression for 10 weeks with GnRH analog [6]. …”

“…Reduced testosterone levels are associated with increased circulating levels of leptin and insulin, even in the absence of increased BMI (Luukkaa et al, 1998;Pitteloud et al, 2005). The hypothalamic-pituitary-gonadal (HPG) axis, which regulates testosterone production, is altered in HD (Bird, Chiappa, & Fink, 1976;Kalliolia et al, 2015;Markianos, Panas, Kalfakis, & Vassilopoulos, 2005;Papalexi et al, 2005;Petersen & Bjorkqvist, 2006;Saleh et al, 2009; Soylu-Kucharz, Baldo, & Petersen, 2016;Van Raamsdonk et al, 2007).…”
