Investigating the impact of internal standard heterogeneity on gold quantification with total reflection X‐ray fluorescence: A simulation study

Abstract: Total reflection X‐ray Fluorescence (TXRF) is a powerful analytical tool with high detection sensitivity that has been applied to a variety of biological samples. While its ability to quantify gold nanoparticles (AuNPs) in cancer cells has been demonstrated, the extension to tissue slices would be of interest. To that end, the preservation of the underlying tissue microstructure requires samples to be measured as microtome slices. In this form, internal standard spiking is warranted. Thus, it is important to e… Show more

“…In order to study the impact of internal standard distribution on quantification, a simulation approach based on TOol for PArticle Simulation (TOPAS) was employed [192]. This model was introduced in a concurrent publication by our group [218]. Briefly, the simulation allows the user to customize the components of a TXRF spectrometer to study the relevant interactions between X-rays and samples of various geometries.…”

“…Sample inhomogeneity can have a major influence on the quality of TXRF spectrometry results. Mankovskii and Pejovic-Milic 60 modelled its influence on the direct analysis of microtome thin slices. They used a profile-fitting based software (TOPAS) to simulate the TXRF instrument and a Python code to simulate the samples.…”

“…Moreover, and in the pursuit of tackling cancer burden, three papers were also selected for this Special Issue. Figueroa et al 7 presented the OXIRIS project, making use of a XRF device for cancer detection and Mankowskii and Pejovi‐Milic 8,9 established a new methodology making use of TXRF for the investigation of gold nanoparticles in cancer treatment.…”
