This study evaluated the role of quercetin against cyclophosphamide‐induced distortion of rat testicular function. Adult rats were administered cyclophosphamide (100 mg/kg), quercetin (50 mg/kg) and in combination for seven days. Cyclophosphamide caused a significant increase in the activities of indoleamine 2, 3‐dioxygenases (IDO), tryptophan 2, 3‐dioxygenase (TDO), myeloperoxidase (MPO), and elevated the concentrations of interleukin 6 (IL‐6) and interferon‐γ (IFN‐γ). Cyclophosphamide increased testis malondialdehyde (MDA) concentrations but depleted superoxide dismutase (SOD), catalase (CAT), glutathione peroxidase (GSH‐Px) and glutathione (GSH). However, quercetin co‐administration significantly (p < 0.05) prevented the increased values of IDO, TDO, MPO, IL‐6, IFN‐γ, MDA, SOD, CAT, GSH‐Px and GSH compared with control rats. Also, quercetin co‐treatment significantly increased serum testosterone, follicle‐stimulating hormone (FSH), prolactin, luteinizing hormone (LH), activities of testicular 3β‐hydroxysteroid dehydrogenase (3 β‐HSD), 17β‐hydroxysteroid dehydrogenase (17 β‐HSD) as well as sperm count, motility and viability but reduced abnormal sperm morphology. Quercetin exposure alone did not alter any of the parameters evaluated relative to control. Thus, quercetin protected the testes against cyclophosphamide‐induced alterations in immunosuppressive IDO/TDO activities elicited by oxidative‐inflammatory mediators.

The present study aims to investigate the lung-protective and antiapoptotic effects of Hypericum triquetrifolium Turra (HTT) against cyclophosphamide (CP)-induced lung injury in rats. Thirty-five Sprague Dawley rats were categorized into 5 groups, each consisting of seven members. Phenolic acid and flavonoid contents of this plant were determined. The lung tissue samples cultivated from the rats were examined both histopathological and immunohistochemically for the apoptosis markers of Caspase-3, Bax, and Bcl-2. Histopathological results indicated that structural defects, bleeding areas, and edema had occurred in the lungs of the CP-Alone Group. Besides, Caspase-3 and Bax positivity of the lung cells had also increased while Bcl-2 positivity had decreased. On the other hand, in the HTT + CP Group, HTT was shown to have reversed the aforementioned changes positively. In addition, our in-vivo results were confirmed by the in-silico studies. The changes that occurred in the binding of CP to the active-site amino acid residues of Caspase-3, Bax, and Bcl-2 upon the addition of Hyperoside besides the changes that occurred in their tendency to form hydrogen bonds were accounted for by in silico studies. Based on in vivo and in silico results, HTT could be a strong protective candidate for CP-induced lung injury and apoptosis.

Background and AimChemotherapy, particularly with methotrexate (MTX), often elicits testicular toxicity, leading to impaired spermatogenesis and hormone imbalances. This study aimed to investigate the potential protective effects of selenium (Se) against MTX‐induced testicular injury.Materials and MethodsMale mice were divided into control, MTX, Se, and MTX + Se groups. Histopathological examination involved the preparation of testicular tissue sections using the Johnsen's tubular biopsy score (JTBS) for spermatogenesis evaluation. Biochemical tests included the assessment of testosterone, malondialdehyde (MDA), luteinizing hormone (LH), and follicle‐stimulating hormone (FSH) levels. Real‐time quantitative polymerase chain reaction (RT‐qPCR) was employed to analyze the expression of caspase 3 (casp3), tumor protein 53 (p53), B‐cell lymphoma 2 (Bcl2), and Bcl2‐associated X protein (Bax) genes. Statistical analysis was performed using ANOVA and Tukey's tests (p < .05).ResultsHistopathological analysis revealed significant testicular damage in the MTX group, with decreased spermatogenesis and Leydig cell count, while Se administration mitigated these effects, preserving the structural integrity of the reproductive epithelium. Biochemical analysis demonstrated that MTX led to elevated malondialdehyde (MDA) levels and reduced testosterone, LH, and FSH levels, suggesting oxidative stress and Leydig cell dysfunction. Gene expression analysis indicated that MTX upregulated proapoptotic genes (casp3, p53, and bax) while downregulating the antiapoptotic Bcl2 gene. In contrast, Se treatment reversed these trends, highlighting its potential antiapoptotic properties.ConclusionOur findings underscore the potential of Se as a therapeutic agent to mitigate the reproductive toxicity associated with MTX‐induced testicular injury. Se exerts protective effects by regulating oxidative stress, preserving hormone balance, and modulating apoptotic pathways. These results suggest that Se supplementation could be a promising strategy to alleviate chemotherapy‐induced testicular damage and preserve male fertility.
