Investigation of Laser-Induced Choroidal Neovascularization in the Rat

Abstract: In the rat model of laser-induced neovascularization, newly developed endothelial cells originate from the retina and the choroid. Whereas ring-like surrounding vessels come from the retina, flat endothelial cells in deeper layers are of choroidal origin or may originate from circulating endothelial precursor cells. Dextran angiography has to be regarded critically for visualizing the choriocapillaris and CNV in laser scars. PECAM-1 immunohistochemistry is best for detection and quantification of neovasculariz… Show more

“…Current animal models of CNV mostly use laser injury to fracture Bruch's membrane (37) or use viral transfection of angiogenic factors into RPE cells to induce CNVs (38). In our model, we used mice deficient in SOD1.…”

“…The perfusion of retinal vasculature with fluorescein isothiocyanate (FITC)-dextran and the preparation of flat mounts were both performed as described by McMenamin 22 and Semkova et al 23 An incision was made in the left ventricle and a cannula was inserted, and the descending aorta was then occluded with a clamp. A constant flow pump (Harvard Apparatus, Holliston, MA) was utilized to perfuse the vasculature.…”

“…FITC-dextran perfusion was developed to visualize retinal vessels in the beginning 25 and, hereafter, has been used widely for evaluating entirely developed CNV lesions. 19,26,27 This technique does not allow consistent visualization of vasculature during the early phase of CNV formation, 19,26 and so may provide variable results when used to measure laser-induced CNV. 26 On the other hand, BS isolectin B4 labeling allows the visualization of the formation of laserinduced CNV from the beginning.…”
