Investigation of Molecular Weight, Polymer Concentration and Process Parameters Factors on the Sustained Release of the Anti-Multiple-Sclerosis Agent Teriflunomide from Poly(ε-caprolactone) Electrospun Nanofibrous Matrices

Abstract: In the current work, a series of PCL polyesters with different molecular weights was synthesized and used for the fabrication of nanofibrous patches via electrospinning, as sustained release matrices for leflunomide’s active metabolite, teriflunomide (TFL). The electrospinning conditions for each sample were optimized and it was found that only one material with high Mn (71,000) was able to produce structures with distinct fibers devoid of the presence of beads. The successful preparation of the fibers was det… Show more

“…The incorporation of the extract in the fibers was corroborated since the transmittance band of the carbonyl characteristic of PCL (C=O) at 1719 cm -1 had a slight shift to higher values of 1723 cm -1 for PCL_5.3 and 1726 cm -1 for the sample with 10.6 wt. % (PCL_10.6), which can be attributed to the formation of intermolecular hydrogen bonds between PCL and the P. major extract [35,65].…”

“…Furthermore, the surface erosion of the PCL matrix causes the fibers’ surface to become rough. According to this finding, the breakdown process begins at the PCL fibers’ surface and moves inside the particles [ 50 , 51 ]. This may indicate re-crystallization of the guest polymer [ 52 ].…”

“…This initial burst release is unavoidable [ 91 ]. Consequently, the biphasic release rate of drug-loaded nanofibers is influenced by factors such as the polymer molecular weight [ 92 ], concentration [ 93 ], sheath thickness [ 94 ], additives, and more. For instance, introducing graphene oxide can enhance the mechanical properties of electrospun membranes and slow the release of model drugs by reducing the matrix swelling [ 95 ].…”
