This work aims at examining the influence of lorazepam, an anxiolytic drug with sedative effects, on brain activity and specifically on EEG Fractal Dimension (FD). The main objective is to clarify the reasons for FD increase after drug intake and to establish a relationship between FD and EEG energy bands. 14 healthy subjects that received either 2.5mg of lorazepam (verum case) or placebo (placebo case) participated in the experiment. 20 EEG channels have been used. One-way ANOVA test revealed that lorazepam increases significantly both FD (p = 0) and beta energy band (p = 1.18E-013 for beta1 band and p = 2.29E-011 for beta2 band) and decreases alpha energy band (p = 0.05 for alpha1 and p = 0.0036 for alpha2), whereas there was not any significant difference on placebo subjects before and after drug intake. Moreover, correlation results indicate that there is a strong correlation between FD and beta energy band (mean correlation coefficient = 0.4120 for beta1 and 0.5358 for beta2) and a negative correlation between FD and alpha1 energy band (mean correlation coefficient -0.4930). Additionally, the mean correlation coefficient between FD and a combination of the different energy bands (beta1+beta2-alpha1) is 0.6185 and reaches value 0.684 for channels F7, T6, P4. These results indicate that there is a relationship between EEG energy bands and FD and provide a link between the classic spectral analysis and the complexity analysis.

BackgroundKetamine is swiftly effective in a range of neurotic disorders that are resistant to conventional antidepressant and anxiolytic drugs. The neural basis for its therapeutic action is unknown. Here we report the effects of ketamine on the EEG of patients with treatment-resistant generalized anxiety and social anxiety disorders.MethodsTwelve patients with refractory DSM-IV generalized anxiety disorder and/or social anxiety disorder provided EEG during 10 minutes of relaxation before and 2 hours after receiving double-blind drug administration. Three ascending ketamine dose levels (0.25, 0.5, and 1 mg/kg) and midazolam (0.01 mg/kg) were given at 1-week intervals to each patient, with the midazolam counterbalanced in dosing position across patients. Anxiety was assessed pre- and postdose with the Fear Questionnaire and HAM-A.ResultsKetamine dose-dependently improved Fear Questionnaire but not HAM-A scores, decreased EEG power most at low (delta) frequency, and increased it most at high (gamma) frequency. Only the decrease in medium-low (theta) frequency at right frontal sites predicted the effect of ketamine on the Fear Questionnaire. Ketamine produced no improvement in Higuchi’s fractal dimension at any dose or systematic changes in frontal alpha asymmetry.ConclusionsKetamine may achieve its effects on treatment-resistant generalized anxiety disorder and social anxiety disorder through related mechanisms to the common reduction by conventional anxiolytic drugs in right frontal theta. However, in the current study midazolam did not have such an effect, and it remains to be determined whether, unlike conventional anxiolytics, ketamine changes right frontal theta when it is effective in treatment-resistant depression.

The rise of mathematical developments in the theories of consciousness has led to new measures to detect consciousness in a system. The Integrated Information Theory (IIT) is one of the best mathematical rooted attempts to quantify the level of consciousness in a system with Φ as the effective information generated in a system above its parts. Recently, the IIT has inspired the Perturbational Complexity Index (PCI) to detect conscious states in patients with disorders of consciousness, and it has shown to have almost perfect classification accuracy. In this study, we explore the statistical correspondence between the theoretical Φ and the experimental PCI through a neurocomputational model of coupled oscillators that can be artificially perturbed, which mainly focuses on the dynamics of collective synchronization between subsets of brain areas. Our results reveal that both measures are statistically related but, in principle, this relationship is far to be perfect. These results are discussed in the context of the model of coupled oscillators, which mainly focuses on the dynamics of collective synchronization between subsets of brain areas.
