Involvement of B1 and B2 receptors in bradykinin‐induced rat paw oedema

Abstract: 1 The mechanisms involved in bradykinin (BK)-induced oedema in the rat paw as well as the interactions between BK and several inflammatory mediators, have been investigated. 2 Intraplantar injection of BK (1 nmol/paw) in rats pretreated with captopril (5 mg kg-', s.c.) caused a small amount of oedema formation (0.17 ± 0.05 ml). Des-Arg9-BK (DABK, a selective B, receptor agonist) up to 300 nmol/paw caused minimal oedema (0.03 ± 0.01 ml). 3 Co-administration of prostaglandin E2 (PGE2), prostaglandin 12 (PGI2), c… Show more

“…One possible explanation for these apparent contradictory results could be that the activation of B 2 Rs was having two major effects in the system-an effect on tissue cells inducing the expression B 1 Rs and a protective effect. As for the former effect, there is evidence to suggest that B 2 R activation takes place at the beginning of the inflammatory process, and that via autoregulatory mechanisms, the desensitization of B 2 Rs occurs, followed by the up-regulations of B 1 Rs (23,44). In our experiments, blockade of B 2 Rs with HOE 140 prevented the up-regulation of B 1 Rs in the intestine and lungs of reperfused animals.…”

“…The contralateral paw received 0.1 mL PBS and was used as a control. Oedema was measured by the use of a plethysmometer (Ugo Basile) at several time points (0.5, 1, 2 and 4 h for Cg and 10, 20, 30, 60 and 120 min for BK) following the injection of inflammatory mediators as described previously (Campos and Calixto, 1995;Campos et al, 1996). Oedema is expressed in mL as the difference between the test and control paws.…”
