Involvement of FAK/Src complex in the processes of Escherichia coli phagocytosis by insect hemocytes

Abstract: Recently we demonstrated that lipopolysaccharide promotes activation of the Ras/mitogen-activated protein cascade in hemocytes and that phagocytosis of Escherichia coli by insect hemocytes is mediated by an integrin-dependent process [Foukas et al. (1998) J. Biol. Chem. 273, 14813^14818]. Here we report data concerning the focal adhesion kinase (FAK) tyrosine phosphorylation status in hemocytes in response to E. coli. We demonstrate that E. coli-triggering stimulates a significant increase in tyrosine phosphor… Show more

“…On the other hand, the binding of Trol seemed to activate integrin and increase the level of FAK phosphorylation. The requirement of FAK has been shown for mammals in the integrin ␣ v ␤ 5 -mediated phagocytosis of apoptotic cells (43) or spent photoreceptor outer segment fragments (44) and for insects in the integrin-mediated phagocytosis of bacteria (45). Therefore, it is possible that phosphorylated FAK is included in a signaling pathway activated by integrin ␤, although INA-1 does not seem to need FAK to induce phagocytosis (17).…”

“…In insect hemocytes, antibody-mediated inhibition of Src and FAK has been shown to abolish FAK phosphorylation and phagocytosis of E. coli (38). In retina, daily ␣v␤5 integrin-dependent phagocytosis of spent photoreceptor outer segment fragments by the retinal pigment epithelium was crucial for retinal function, and FAK was pivotal in this process (39).…”

“…This discrepancy is likely to originate from the use of only the Triton X-100 soluble cell lysate fractions in the prior study. Invasion by fimbriated P. gingivalis thus evokes the formation of integrin-associated focal adhesions and will likely stimulate the cascade of events that normally follows integrin activation (Alrutz & Isberg, 1998;Metheniti et al, 2001;Ozeri et al, 2001;McGee et al, 2003).…”
