Involvement of Gpr125 in the myeloid sarcoma formation induced by cooperatingMLL/AF10(OM-LZ)and oncogenicKRASin a mouse bone marrow transplantation model

Abstract: Oncogenic N-/KRAS mutations were frequently associated with MLL/AF10 in acute myeloid leukemia with myeloid sarcoma (MS). To study the cooperating leukemogenesis by MLL/AF10 and KRAS mutation, we retrovirally transduced MLL/AF10(OM-LZ) and KRAS G12C into mouse bone marrow cells and generated two Chromosomal translocations involving MLL gene at chromosome 11q23 were detected in patients with de novo acute leukemia (AL) and therapy-related acute myeloid leukemia. 1More than 60 MLL translocation partners have bee… Show more

“…Knocking down Gpr125 in murine myeloid sarcoma lines that harbor KRAS G12C and MLL/AF10(OM-LZ) oncogenes leads to reduced myeloid sarcoma burden in mice, and a decrease in cell adhesion in culture, possibly contributing to their reduced tumorigenicity [106]. …”

“…Thus, downregulation of GPR124 is discussed as a therapeutic approach to overcome NSCLC gefitinib resistance [127]. Knockdown of Gpr125 reduces tumor cell aggregation and diminishes myeloid sarcoma formation induced by transplantation of immortalized acute myeloid leukemia [106]. …”

“…Two studies use the same MSCV based BM transduction with MLL-AF10 and both models develop myeloid lineage disease (74, 75). An additional model studied the cooperativity of MLL-AF10 with the activating mutation Kras G12C (76). …”

“…In this disease, which co-presents with acute myeloid leukemia, myeloid cells accumulate in peripheral tissues as a solid mass, atypical of blood borne tumors. In their study, Fu et al (Fu et al, 2013) showed that Gpr125, which is not typically expressed in hematopoietic lineages (Haitina et al, 2008), was upregulated in MS cells and that RNAi against Gpr125 decreased the adherent abilities of MS cells ex vivo . The mechanism for this observation is still unclear.…”
