Involvement of lipids in ferriprotoporphyrin IX polymerization in malaria

Abstract: Approximately 70% of the initial ferriprotoporphyrin IX polymerizing activity in cell-free preparations of erythrocytes infected with Plasmodium berghei was recovered in a chloroform extract. No polymerizing activity remained in the residue. In studies to identify substances that promote FP polymerization, arachidonic, linoleic, oleic, and palmitoleic acids, 1-mono- and di-oleoylglycerol, and the detergents, SDS, Tween 80, and n-octyl-glucopyranoside, were active. Tri-oleoylglycerol, cholesterol, di-oleoylphos… Show more

“…Next, we examined whether aptamers interfere with hemozoin formation in vitro using 1-mono-oleoylglycerol (MOG) (30) as an initiator. In these assays, MOG and heme were co-incubated in pH 5.2 buffer and the nucleic acid concentration titrated to attain the desired final concentration.…”

“…among these are the family of unusual histidine rich protein, HRP I-IV, (Sullivan et al, 1996a;Choi et al, 1998)which bind large quantities of heme. In addition, glycoproteins and lipids have been suggested to mediate malaria pigment formation (Arese et al, 1999;Fitch et al, 1999). Clearly the reactions outlined in equation 1 and 2 can generate aggregated heme phases without catalysis, but the role of macromolecular catalysis of these reactions remains an important research effort.…”

“…This raised the possibility of oriented nucleation, although the clustering and alignment might have occurred after the nucleation step. Fitch et al (2) have proposed that neutral lipids play a role in inducing crystallization of hemozoin within the digestive vacuole (DV). The presence of lipid bodies a few hundred nm in diameter, composed of mono-and diacylglycerols close to the outer surface of the DV, has been reported by Jackson et al (3).…”
