Involvement of NF-κB/IL-6 Pathway in the Processing of Colorectal Carcinogenesis in Colitis Mice

Abstract: Nuclear factor-kappaB (NF-κB)/interleukin (IL-6) pathway links chronic inflammation to colitis associated cancer (CAC). In this study, we examined the dynamic temporal changes of the NF-κB/IL-6 pathway during the procession of experimental CAC mouse model. Mice were sacrificed after induction for 14, 16, 18, and 22 weeks for the examination of tumor burden, inflammation degree, and protein level of NF-κB and IL-6 in bowel tissues. The results showed that tumor burden and inflammation severity in the bowels wer… Show more

“…At the same time, populations that are continuously exposed to As may chronically produce IL-8 as a result of the increases in the NF-κB mRNA levels [27]. The IL-6 and IL-8 mRNA expression by the liver tissue was significantly increased with the stimulus [28][29][30][31]. Our results on these endpoints are not broadly in agreement with our previous measurements of cytokines induced by toxins.…”

“…The abnormal activation of NF-κB contributes to significant cell proliferation and migration in CRC and in other types of cancer ( 3 ), and the inhibition of NF-κB activity has been shown to significantly reduce cancer cell growth and enhance cell apoptosis, indicating that NF-κB members are potential therapeutic targets in tumors ( 4 ). Mammalian NF-κB members mainly include p65 (RelA), p105/p50, RelB, p100/p52 and c-Rel ( 5 ), and two distinct pathways are involved in the regulation of NF-κB activation: the canonical and the non-canonical pathways. The canonical pathway of NF-κB is controlled by the IκB kinase (IKK) complex, which is comprised of IKKα, IKKβ and IKKγ.…”

“…Previous reports have found DSS increases LCN2 in both plasma and feces [312,314], while LCN2 mRNA was not induced in WT mice treated with paraquat [315]. Similarly we saw increased IL-6 following LPS and paraquat administration, as we and others have previously reported [298,316], however, no effect of DSS was observed on this measure contrary to previous reports in the gut [317]. In addition, we further found no alteration in IL-1B or IL-10 with either DSS or LPS and paraquat treatment.…”
