Involvement of NF-κB-mediated expression of galectin-3-binding protein in TNF-α-induced breast cancer cell adhesion

Abstract: Abstract. Galectin-3-binding protein (G3BP) is highly expressed in various types of cancer and is thought to be involved in cancer malignancy; however, the role of G3BP in breast cancer cells is not fully understood. In this study, we investigated the role of NF-κB in the adhesion of breast cancer cells to a substrate by using (-)-DHMEQ, a specific inhibitor of NF-κB. (-)-DHMEQ inhibited both TNF-α-induced G3BP expression and cell adhesion in human breast cancer cell lines. We also found that knockdown of G3BP… Show more

“…Gal-3 has been considered a tumor-associated antigen of malignancy level in breast cancer, and is implicated in aspects, such as angiogenesis [219], adhesion [220], invasion [221], apoptosis inhibition [222], migration [223], and therapeutic resistance [224]. Chen et al, through hydrazide-based chemistry method combined with liquid chromatography tandem-mass spectrometry (LC-MS/MS), identified 72 unique N-linked glycoproteins that were significantly regulated in triple negative breast cancers, among which is LGALS3BP.…”

“…(1) Galectin-3-binding protein (LG3BP) was reported to be a candidate diagnostic biomarker for large-cell neuroendocrine lung carcinoma 26 . In addition, galectin-3-binding protein was also reported to induce galectin-mediated tumor cell aggregation to increase the survival of cancer cells in the bloodstream during the metastatic process 27,28 . (2) Alpha-1-acid glycoprotein (A1AG) was reported as potential lung cancer serum biomarker 29 .…”

“…It is a binding partner of Gal-1 and Gal-3, which promote integrin-mediated cell adhesion, and significantly elevated expression of Gal-3 BP in the serum or tumor tissues is associated with poor prognosis in a variety of malignancies including breast cancer (29), lung cancer (30), and PC (31). Additionally, Gal-3 BP is involved in the promotion of integrinmediated tumor cell adhesion to the ECM proteins in colon cancer (32), breast cancer (33), and the formation of metastasis in lung cancer (34). Our study reveals that targeting Gal-3 BP may prevent the stimulatory effect of SCs in PC cancer cells.…”
