Involvement of plasma adipokines in metabolic and reproductive parameters in Holstein dairy cows fed with diets with differing energy levels

Abstract: This study aimed to investigate the association between plasma adipokine concentrations and metabolic and reproductive parameters in Holstein dairy cows fed diets with different energy levels during the peripartum period. The experiment started 1 mo before first calving and was maintained for 2 lactations. Dry matter intake and energy balance in animals fed a low-energy (LE) diet were significantly lower than that of animals fed a high-energy (HE) diet in the first lactation. Body weight, milk production, back… Show more

“…During the bovine oestrus cycle, serum adiponectin levels range between 27 and 32 μg/ml and were 1. follicular fluid, but the concentrations in the serum and follicular fluid were not correlated [33]. The circulating levels of adiponectin were decreased when providing low energy diet to the cows, and were related to a delayed and abnormal luteal activity in lactating cows [34]. In vitro, adiponectin (3 μg/ml) decreases the production of androstenedione (A4) by TC in cows by reducing the expression of LH receptors and CYP11A1 (cytochrome P450, family 11, subfamily a, polypeptide 1) and CYP17α1 (cytochrome P450, family 17, subfamily a, polypeptide 1) enzymes mediated by both receptors AdipoR1 and AdipoR2.…”

“…These findings are in agreement with Dubuc et al (2011) and reinforce the subclinical nature of this condition. In dairy cows, plasma concentrations of adipokines are affected by negative energy balance status ( Mellouk et al 2017 ), variations in white adipose tissue quantities ( Reverchon et al 2014 ) and milk yield ( Kafi et al 2015 ). However, no differences were observed here between uterine health groups regarding these parameters.…”

“…Plasma and uterine fluid total ADIPOQ, RARRES2 and NAMPT concentrations were measured by using ELISA kits (E11A0125 and E11C0104 from BlueGene, Shanghai, China and EK-003-80 from Phoenix France, SAS, Strasbourg, France, respectively), as previously described ( Fadden & Bobe 2016 , Mellouk et al 2017 , 2019 ). The intra-assay coefficients of variation for total ADIPOQ, RARRES2 and NAMPT were 6%, 4.5% and 6.5%, respectively.…”

“…pp. The composition of the HE and LE diets was previously described [25] (Additional file 1: Table S1). The energy content of the diet was about 1.16-fold higher in the HE than in the LE diet (84.23 vs. 72.35 Mcal/kg of dry matter).…”

“…The extent and duration of NEB may lead to some reproductive dysfunctions such as a reduced pulsatile LH release [23], and ovulatory failures [24]. These events are associated with modifications in adipokines (resistin, adiponectin and leptin) blood/serum or plasma concentrations, which are regulated by the energy level of the diet in dairy cows [25].…”
