Ionic currents expressed in a cell line derived from the organ of Corti of the Immortomouse

Abstract: We have investigated the maturation of adult hair cell electrophysiology in a population of precursor cells in a conditionally immortal cell line. The cell line, UB/OC-2, from the embryonic organ of Corti of the H-2Kb-tsA58 transgenic mouse, permits cells to grow proliferatively at 33 degrees C and to differentiate at 39 degrees C. Whole-cell patch-clamp recordings showed that proliferating cells had a different electrophysiology to differentiating cells. Differentiating cells had a conditionally expressed slo… Show more

“…Following application of acetylcholine the inward currents were clearly visible because the outward, calcium-activated potassium conductance was absent (see Evans, 1996), consistent with the fact that it is normally expressed a few days later (Dulon and Lenoir, 1996). There is evidence for expression of the outward current after longer periods in culture (Jagger et al, 1999). This suggests that the sequence of gene expression in UB/OC-1 is similar to that observed in vitro although the time scale is longer.…”

“…The currents recorded under proliferating culture conditions and the I K,early current are similar to those recorded from other cochlear epithelial cell lines (Jagger et al, 1999;Lawlor et al, 1999). The reversal potentials suggest that they are mainly carried by K + ions.…”

“…This would allow us to ascertain the time course of development of K + currents in OHCs and IHCs. The results should help us to link the expression of physiologically important hair cell genes to the age-dependent expression of specific regulatory genes and to interpret findings from emerging studies on conditionally immortal cell lines Jagger et al, 1999].…”
