Ionization of anabolic steroids by adduct formation in liquid chromatography electrospray mass spectrometry

Abstract: The ionization of 46 anabolic steroids has been studied. The absence of basic or acidic moieties in most of these analytes makes their direct ionization as [M + H]+ by atmospheric pressure interfaces difficult. The formation of adducts with different components of the mobile phase has been found to be an efficient way to ionize anabolic steroids by electrospray. Different mobile phases using methanol (MeOH) or acetonitrile as organic solvent and HCOOH, Na+ or NH4+ as additives have been tested to favor the add… Show more

“…After metabolic activation of DHEA with bovine liver S9 and cofactor NAD (Fig. 5) the main metabolites of DHEA (X) appeared to be 4-adione + ions, whereas the base peak in the mass spectra of 4-adione and 17β-T was the [M+H] + ion, as expected for 3-keto-4-ene steroids [21].…”

“…The ionization of A2 and A3 as [M ϩ H] ϩ by atmospheric pressure interfaces is difficult owing to the absence of a keto function (Pozo et al, 2007c). Therefore, the detection of 17␣-methyltestosterone misuse is not possible by current methods based on liquid chromatographytandem mass spectrometry (LC-MS/MS) (Mazzarino and Botrè, 2006;Pozo et al, 2007b).…”

“…pdf). However, the unequivocal proof of 17␣-methyltestosterone misuse is difficult because both A2 and/or A3 are also metabolites of other anabolic steroids including metandienone, mestanolone, methandriol, or oxymetholone (Schänzer and Donike, 1993).The ionization of A2 and A3 as [M ϩ H] ϩ by atmospheric pressure interfaces is difficult owing to the absence of a keto function (Pozo et al, 2007c). “…When using an LC mobile phase of methanol/water/ammonium acetate (1 mM) and 0.001 % acetic acid, Pozo et al [39] reported the formation of [M+CH 3 COO] -for several anabolic steroids. Antignac et al [40] observed [M+ CH 3 COO] -formation from corticosteroids in biological matrices of cattle when employing a methanol/water/1 % acetic acid mobile phase.…”
