Ionotropic glutamate receptors activate cell signaling in response to glutamate in Schwann cells

Abstract: In the peripheral nervous system, Schwann cells (SCs) demonstrate surveillance activity, detecting injury and undergoing -differentiation to support repair. SC receptors that detect peripheral nervous system injury remain incompletely understood. We used RT-PCR to profile ionotropic glutamate receptor expression in cultured SCs. We identified subunits required for assembly of-methyl-d-aspartic acid (NMDA) receptors (NMDA-Rs), α-amino-3-hydroxy-5-methyl-4-isoxazolepropionic acid receptors, and kainate receptors… Show more

“…Finally, riluzole can protect against mitochondrial dysfunction and oxidative stress (Shimizu et al 2018). The regulation of PKC and AMP-activated protein kinase signals (Noh et al 2000;Daniel et al 2013;Mehnert et al 2018), induced by the overactivation of glutamate receptors or ion channels (Campana et al 2017;Aromolaran et al 2018), may represent a mechanism of action by which riluzole reduces oxidative stress (Cai et al 2017;Guo et al 2018).…”

“…Phosphorylated proteins were detected in an unbiased manner using the human Phospho-kinase Array Proteome Profiler TM (R&D System). Although this system was originally developed to examine human proteins, we have demonstrated its effectiveness using rodent cell extracts (21,64). Microglia in 0.5% serum-supplemented medium were treated with RAP (150 nM) or vehicle (PBS) for 1 h. Protein extracts were prepared and applied to the membranes.…”
