Irisin Ameliorates PM2.5-Induced Acute Lung Injury by Regulation of Autophagy Through AMPK/mTOR Pathway

Abstract: Background: PM2.5 exposure is one of the major inducements of various respiratory diseases and related mortality. Meanwhile, irisin, a metabolism and thermogenesis-related hormone, is found to be protective against acute lung injury induced by LPS, which indicates its therapeutic function in lung injury. However, the function and underlying mechanism of irisin in PM2.5-induced acute lung injury (ALI) are still unclear. This study is aimed to discover the potential mechanisms of irisin in PM2.5-induced acute lu… Show more

“…Additionally, the antioxidative pathway had a positive and direct impact on autophagy. In fact, silencing Sirt1 impaired autophagy in the Ad-mCherry-GFP-LC3B transfection assay, with both GFP green and mCherry red fluorescence becoming yellow puncta [48].…”

“…Autophagy can promote phagocytosis and antigen presentation by macrophages (32) and inhibit macrophage M1-type polarization, which prevents imbalances in the in ammatory response (33). Autophagy can also induce metabolic reprogramming in type II alveolar epithelial cells (AT2) by converting lipid metabolism to glucose metabolism, thus promoting the proliferation of AT2 cells, which facilitates pulmonary homeostasis and injury repair (34,35). Therefore, autophagy plays a crucial role in maintaining metabolic homeostasis and in the development and pathogenesis of ALI.…”

“…PM 2.5 was collected using a multi-level flow particulate sampler (2030, Laoying Haina Photovoltaic Environmental Protection Group Co., Qingdao, China) from a dairy cattle farm in Changchun, Jilin Province [ 36 , 37 , 38 ]. The sampler was located at the center of the cow farm, 1.5 m above the ground, with a sampling flow rate of 100 L/min that operated continuously for 24 h. After collection, ultra-pure water without endotoxin was used for elution, which was then filtered through six layers of sterile gauze and freeze-dried using a vacuum lyophilizer.…”
