IRX5 promotes colorectal cancer metastasis by negatively regulating the core components of the RHOA pathway

Abstract: Colorectal cancer (CRC) is one of the most common malignant tumors worldwide. As tumor metastasis is the leading cause of death in patients with CRC, it is important to elucidate the molecular mechanisms that drive CRC metastasis. Studies have shown a close relationship between Iroquois homeobox (IRX) family genes and multiple cancers, while the mechanism by which IRX5 promotes CRC metastasis is unclear. Therefore, we focused on the involvement of IRX5 in CRC metastasis. In this study, analyses of clinical dat… Show more

“…IRX5 alone has also been reported to be an oncogenic gene in hepatocellular carcinoma, colorectal cancer, prostate cancer, and non-small cell lung cancer by regulating cancer cell cycle and apoptosis (Myrthue et al, 2008;Zhang D.L. et al, 2018;Zhu et al, 2019Zhu et al, , 2020. Although there is no direct evidence that FTO SNPs are associated with cancer by affecting the expression of adjacent genes, we cannot rule it out as a possibility.…”

“…The IRX5 is a transcription factor that is closely associated with a variety of malignancies 29,30 . IRX5 can promote the invasion and migration of colorectal cancer cells by inhibiting the RHOA‐ROCK1‐LIMK1 axis 31 . IRX5 expression has been shown to be positively correlated with OS in smokers and negatively correlated with OS in non‐smokers with LUAD 32 .…”

“…Recently, RHOA is frequently reported as an oncogenic gene implicating in the initiation and progression of malignant tumors via exacerbating cell migration and invasion [6,8,19,20]. RHOA is widespreadly overexpressed in prostate cancer, cervical cancer and colorectal cancer, and associated with cancer metastasis [21][22][23].…”
