Is non-buffered DMEM solution a suitable medium for in vitro bioactivity tests?

Abstract: Several laboratories had tested bioactivity of the materials in commercially available solution DMEM (Dulbecco's Modified Eagle's Medium) that is normally used for cultivation of cell cultures. The objective of this work was to find out whether it is possible to replace TRIS-buffered SBF currently used for bioactivity tests with the non-buffered DMEM solution. To understand the role of the organic part of the DMEM solution in the process of crystallization, we have prepared non-buffered solution simulating onl… Show more

“…EBSS contains inorganic salts (NaCl, KCl, CaCl 2 , MgSO 4 , NaHCO 3 , NaH 2 PO 4 ) and was successfully used for CaPO 4 crystallization (Termine and Eanes 1974). Other popular physiological solutions comprise Hanks' balanced salt solution (HBSS), which contains almost similar inorganic salts and glucose (Hanks and Wallace 1949;Shibata et al 2004;Marques et al 2003a;Mareci et al 2010), Eagle's minimum essential medium (MEM) and its variation Dulbecco's modified Eagle's medium (DMEM), which contain numerous bioorganic (alanine, aspartic acid, glycine, biotin, vitamin C, folic acid, riboflavin) and inorganic (CaCl 2 , KCl, NaCl, NaH 2 PO 4 ) components (Meuleman et al 2006;Touny et al 2011;Coelho et al 2000;Mandel and Tas 2010;Rohanová et al 2014), phosphate-buffered saline (PBS) that contains only inorganic (CaCl 2 , MgCl 2 , KCl, KH 2 PO 4 , NaCl, NaH 2 PO 4 ) components (Gao et al 2006;Lichtenauer et al 2011). Furthermore, artificial saliva Ionta et al 2014;Okulus et al 2014), synthetic urine (Assimos 2013Dbira et al 2015) and simulated milk ultrafiltrate (SMUF) (Jenness and Koops 1962;Spanos et al 2007;Gao et al 2010a, b) solutions are available.…”

“…Therefore, in the literature, attempts to substitute SBF with the commercially available cell culture media, i.e. HEPES-free Dulbecco's modified Eagle medium (DMEM) solution, have been reported [15,17]. Apart from the inorganic part of blood serum, this solution contains also the organic component (vitamins, amino acids, etc.).…”

“…Moreover, DMEM can be supplemented with foetal bovine serum (FBS), containing additional organic species, such as proteins, to obtain complete growth medium for certain cell lines. The medium can be carbonate buffered as the test is performed in a 37°C incubator under a humidified atmosphere of 95% air and 5% CO 2 , which better simulates the in vivo environment [15,17].…”

“…7a). This phenomenon could be due to the interaction between BGN and DMEM causing an amorphous Ca-P layer formation on the surface of the particles, 51 One approach to ensure continuous ion-release is to incorporate metallic elements into the silicate network. The SiO 4 tetrahedron is the basic building unit of the silicate glasses and the tetrahedron can bond to each other through Si-O-Si bonds.…”

“…50 However, DMEM intrinsically contains abundant levels of potassium, calcium, and amino acids, leading to possible interaction with the tested material over time. 51 In this study, the assessment medium (DMEMWater mixture) contained Ca 2+ ions at a concentration of $6.5 mg mL À1 . It is notable that Ca 2+ ions were released from BGN-30 during the rst 1 hour immersion while the calcium concentration in the medium decreased in the following 3 days of immersion (Fig.…”
