Is there any difference between endometrial hyperplasia and endometrial carcinoma in terms of expression of TRPM2 and TRPM7 ion channels?

Abstract: Background/aim: This study compared TRPM2 and TRPM7 ion channel gene expression and immunohistochemical staining in endometrial hyperplasia and endometrium adenocarcinoma. Materials and methods: Sections were taken from paraffin blocks of 120 patients who were divided into 6 groups as follows: G1 (n = 20), proliferative endometrium (PE); G2 (n = 20), EH without atypia; G3 (n = 20), EH with atypia; G4 (n = 20), stage 1A, grade 1 EC; G5 (n = 20), stage 1A, grade 2 EC; and G6 (n = 20), stage 1A, grade 3 EC. TRPM2… Show more

“…Histoscore was calculated for immunohistochemical staining. Histoscore = distribution (0.1 = under 25%; 0.4 = 26-50%; 0.6 = 50-75%; 0.9 = 76-100%) × intensity (0 = no staining; + 0.5 = very little staining; + 1 = little staining; + 2 = medium staining; + 3 = very strong staining) [21].…”

“…Extensity of the staining was taken as the basis when evaluating the immunohistochemical staining. A histoscore was derived from the distribution (0.1 = under 25%; 0.4 = 26-50%; 0.6 = 50-75%; 0.9 = 76-100%) and the intensity (0 = no staining; +0.5 = very little staining; +1 = little staining; +2 = medium staining; +3 = very strong staining) of the staining immunoreactivity (Histoscore = distribution × intensity) [24].…”

“…Recent reports suggest that the activity of the TRPM7 ion channel may indicate the progression from endometrial hyperplasia to early-stage EC. [2] While early-stage EC cases can often be successfully treated through a hysterectomy, advanced EC poses significant challenges with a bleak prognosis and a lack of reliable markers and effective treatment options. However, advancements in targeted chemotherapeutic approaches, such as immunotherapy targeting immune checkpoints and novel chemotherapy strategies, have emerged based on the molecular understanding of EC.…”
