IS292: a novel insertion element from Agrobacterium

Abstract: A new insertion sequence, 15292, located in the 6b gene of a nopaline-type Agrobacterium strain (X88-292) isolated from poplar was identified and sequenced. IS292 is 2494 bp long, has 21 bp inverted terminal repeats with two mismatches, and generates 10 bp direct repeats upon integration. No sequence similarity was found between IS292 and other insertion elements associated with Agrobacterium, but it shows strong similarity with lSRI7 from Rhizobium leguminosamm bv. viciae. The occurrence of IS292-like sequenc… Show more

“…As is the case with relatively homologous elements found in A. xylinum, S. meliloti and A. tumefaciens (Coucheron 1993;Laberge et al 1995;Ponsonnet et al 1995), ISZm1068 was found to be present in single copy in the genomes of different Z. mobilis strains (Fig. 4), a fact that possibly indicates a lack of mobility inside the native organism.…”

“…Two types of PCR primers were designed making use of (a) homologies between the transposase coding regions, and (b) conserved IR sequences of ISs that belong to the genera of Acetobacter (Coucheron 1993;Iversen et al 1994), Agrobacterium (Machida et al 1984;Bonnard et al 1989;De Meirsman et al 1990;Paulus et al 1991;Wabiko 1992;Ponsonnet et al 1995), Sinorhizobium (Dusha et al 1987;Soto et al 1992;Selbitschka et al 1995) and Pseudomonas (Ferrante and Lessie 1991). Primers that finally resulted in the amplification of single fragments from the Z. mobilis strain CP4 genome were GT2 alone or in combination with either GT11 or GT12.…”

“…1 d) contains additional essential Vir operons from pTiBo542 including virA, virJ, virB, virG, virC, virD and virE , associated with plant transformation (reviewed in (Nester 2014 )). A 3.2 kb fragment including the IS292-like insertion element (Ponsonnet et al 1995 ) between virA and virJ and an 8.1 kb fragment between virJ and virB1 were deleted from the vir gene cluster in plasmid pPHP79761 (Fig. 2 ).…”
