IscR Regulates Synthesis of Colonization Factor Antigen I Fimbriae in Response to Iron Starvation in Enterotoxigenic Escherichia coli

Abstract: Iron availability functions as an environmental cue for enteropathogenic bacteria, signaling arrival within the human host. As enterotoxigenic Escherichia coli (ETEC) is a major cause of human diarrhea, the effect of iron on ETEC virulence factors was evaluated here. ETEC pathogenicity is directly linked to production of fimbrial colonization factors and secretion of heat-labile enterotoxin (LT) and/or heat-stable enterotoxin (ST). Efficient colonization of the small intestine further requires at least the fla… Show more

“…Within this set, only 22 TFs had putatively assigned functions. For these TFs, homologues in other organisms are involved in the regulation of metabolism, detoxification and virulence, suggesting that they may fine tune the expression of essential genes important for basal and conserved physiological processes [49, 53–60]. …”

“…[187,188] Thei nhibition of heme-iron acquisition systems of other bacteria has not yet gained as much interest as the system of S. aureus,a lthough iron-regulated expression of hemolysin is also reported for other bacteria, such as Bacillus cereus, E. coli, and Vibrio cholera. [189,190] One of the rare examples of inhibition of heme uptake is by the structure-guided development of synthetic metal complexes, which replace heme in the binding site of the heme acquisition system A( HasA) protein. HasA is secreted by P. aeruginosa,b inds heme "like apair of tweezers", and transfers the heme to the hemophore-…”
