Isoflurane Facilitates Synaptic NMDA Receptor Endocytosis in Mice Primary Neurons

Abstract: Inhalation anesthetic isoflurane has been reported to induce caspase activation and accumulation of β-amyloid (Aβ), however, the down-stream consequences of these effects are largely unknown. Isoflurane has also been shown to impair learning and memory, however, the up-stream mechanisms of these effects remain largely to be determined. Facilitation of synaptic NMDA receptor endocytosis can reduce synaptic function, leading to learning and memory impairment. We therefore set out to determine the effects of isof… Show more

“…It is known that isoflurane induces suppression effect to neuronal activities via exerting antagonistic actions on N-methyl-d-aspartate receptors and enhancing GABA A receptor-mediated functions (Brosnan, 2011;Dong et al, 2013;Harrison et al, 1993;Shelton and Nicholson, 2010). The suppression effect is dose dependent, as seen in the isoflurane-induced burst suppression pattern (Ferron et al, 2009).…”

“…Sevoflurane was found to increase TNF-alfa and production of beta-amyloid through activation of caspases and apoptotic cascade [19], which are major characteristics of PD [20]. …”

“…The hypnotic agents with apoptotic activity could be classified as two main groups: N-methyl-D-aspartate (NMDA) antagonist, like ketamine; and gamma-aminobutyric acid (GABA) receptor agonists, like propofol or thiopental (45-50). Of course, hypnotic agents and amnestic agents have some overlapped clinical use; but we have discussed them under two different subtitles.…”
