Isolation and Characterization of CD271+ Stem Cells Derived from Sheep Dermal Skin

Abstract: Background: Mesenchymal stem cells (MSCs) have great promise in the field of regenerative medicine due to their differentiation potential into several lineages. Besides the bone marrow, MSCs can be obtained from the dermis, which represents a large stem cell reservoir in the skin. Sheep provide an appropriate large animal model for preclinical studies. In this study, we focused on the isolation and characterization of MSCs from sheep dermis as an alternative to bone marrow MSCs (bmMSCs). Methods: Primary ovine… Show more

“…MSC were isolated from caprine BM using standard method based on the ability to adhere and proliferate on plastic surfaces. Isolated population displayed features typical for mesenchymal stem cells as previously demonstrated in small ruminants (Murphy et al, ; Jahroomishirazi et al, ). Caprine MSC possessed remarkable proliferation capacity.…”

“…Previous studies have reported the isolation of oMSCs from bone marrow [ 5 , 23 25 ], adipose tissue [ 34 36 ], umbilical cord blood [ 36 , 37 ], peripheral blood [ 21 , 38 ], amniotic fluid [ 39 , 40 ], dermis [ 41 ] and periodontal ligament [ 42 ]. Despite the increasing use of sheep as a large animal model for tissue engineering, because of their similarities to humans in size, joint architecture and healing mechanisms [ 25 ], ovine MSCs have been poorly characterized compared to human MSCs [ 24 ].…”

“…Mesenchymal stem cells are characterized by the ability to renew and differentiate both into mesodermal tissues (osteoblasts, adipocytes, chondrocytes and myocytes) and other cell types, such as astrocytes and neurons [1][2][3]. In this regard, the isolation of mesenchymal stem cells from bone marrow [4,5], adipose tissue [6][7][8], umbilical cord blood [9], peripheral blood [10,11], as well as from the amniotic fluid [12] and the dermal tissue [13] have been published.…”
