Isolation and characterization of multiple cell types from a single human colonic carcinoma: Tumourigenicity of these cell types in a xenograft system

Abstract: The HCA-7 cell line was established from a moderately well differentiated mucinous adenocarcinoma of the colon, which showed histological heterogeneity. This was reflected in the morphological heterogeneity in early passages of the HCA-7 cell line but diminished as the cells were passaged in vitro. Nine subpopulations were isolated from early passage cultures of the HCA-7 line and maintained as cell lines. Each subpopulation demonstrated a unique set of stable biological characteristics in vitro. When establis… Show more

“…Transient Transfection Assay. The HCA7 human colon cancer cell line was established from a moderately differentiated adenocarcinoma of the colon (14). The cell line was maintained in DMEM supplemented with 100 IU/mL penicillin, 100 g/mL streptomycin, and 10% FBS.…”

“…Indomethacin and 1 were compared for their ability to activate the nuclear transcription factor PPAR␥ in the human colon cancer cell line HCA-7, which basally expresses this nuclear receptor (14,15) Cells were transfected with an expression vector containing luciferase under the control of a PPRE. Addition of either compound triggered a concentration-dependent increase in luciferase activity ( Figure 2, panel a).…”

“…Three human colorectal cell lines were used in this study: HCA-7 Colony 29 (Marsh et al, 1993) termed Colony 29 in this study, Caco-2 (ECACC) and a twice cloned population of HRA-19 cells (Kirkland, 1988), which have been subjected to multiplex PCR analysis (ECACC; Porton Down, Salisbury, UK) to confirm their unique profile. Cell lines were grown in Dulbecco's Eagle's medium (Invitrogen, Paisley, UK) supplemented with 10% foetal bovine serum (FBS) in 7.5% CO 2 .…”
