Isolation and characterization of Sendai virus DI-RNAs

The porcine paramyxovirus is a newly identified agent of a fatal disease in piglets, endemic in Mexico since 1980 Analysis of nucleocapsids obtained from purified virus or from a permanently infected cell line revealed one major band with an M~ of 68K, the nucleoprotein. Two other proteins were also identified, the large protein and the matrix protein, with apparent Mr of about 200K and 40K, respectively. The protein migration pattern of LPMV was compared, by SDS-PAGE, with that of Newcastle disease virus, bovine parainfluenza 3 virus and Sendai virus. Differences in the Mr of LPMV proteins and the proteins of these paramyxoviruses were observed. We propose that LPMV should be classified as a novel member of the genus Paramyxovirus.

SUMMARYThe fate of the haemagglutinin-neuraminidase glycoprotein (HN) of Sendal virus in three types of infection was studied by measuring its sensitivity to endoglycosidase H and its rate of appearance and turnover at the cell surface. HN behaved differently in the three types of infection. When highly expressed at the surface, as in a lytic standard virus infection, HN accumulated at the surface in a stable form (half-life of disappearance from the surface > 10 h). When moderately expressed, as in a non-lyric standard virus plus defective interfering virus infection, HN reached the membrane normally, but turned over rapidly (half-life about 2 h) and was re-internalized. When poorly expressed, as in long-term persistent infection, HN did not reach the cell surface and appeared to be degraded before reaching it. In contrast to HN, the other viral glycoprotein, F0, exhibited a similar turnover rate at the cell surface in the three situations. However, when compared to surface expression in standard virus-infected cells under standardized conditions, F0 surface expression in persistently infected cells was reduced. This reduction correlates with a decreased maturation rate in these cells.

The Characteristics Required for a Sendai Virus Preparation to Induce High Levels of Interferon in Human Lymphoblastoid Cells

SUMMARYThe capacity of an egg-grown Sendai virus preparation to induce interferon in the human lymphoblastoid cell Namalwa is dependent on its passage history. Virus which has been serially passaged at high dilution is a poor inducer, whereas virus serially passaged undiluted is a good inducer. Such a good inducer preparation has a low infectivity to haemagglutination ratio as the result of a high content of defective-interfering (DI) particles. Using DI particles purified on glycerol gradients, it is shown that for the induction of maximum interferon titres both infectious and DI particles are required. DI particles alone induce little or no interferon. Addition of DI particles to fully infectious Sendai virus preparations increased the interferon yield obtained from Namalwa cells some 60-to 100-fold.
