Isolation and characterization of ten microsatellite loci for the Chong’an moustache toad Leptobrachium liui (Anura: Megophryidae)

Abstract: We isolated and characterized ten polymorphic microsatellite loci in the Chong'an moustache toad Leptobrachium liui, nine novel loci designed for this species and one locus from cross-species amplification. Loci were screened in 43 individuals of a single population. The number of alleles per locus ranged from 3 to 17, on average of 10. The average observed and expected heterozygosity per locus ranged from 0.512 to 0.884 and from 0.580 to 0.898, respectively. No significant departures from Hardy-Weinberg equil… Show more

“…The microsatellite loci included VIB‐B4, VIB‐C10, VIB‐D5, CHA1, CHA6, CHA9, LSMT6, LSMT8, CHA4, CHA7, CHA8, CHA11, LSMT4, and LSMT9 (Bi, Deng, Crosby, & Fu, ; Hu, Xia, Zheng, & Zeng, ; Wang et al., ). The PCR fragments containing primers fluorescently labeled with FAM, TAMRA, or HEX were evaluated along with the GS500 size standard using an ABI 3730 analyzer (Applied Biosystems, Foster City, CA, USA).…”

“…For microsatellites amplification, we used eight loci (LSMT1, LSMT8, LSMT9, CHA1, CHA4, CHA9, VIB-C5 and VIB-D5) that could be amplified in three species (Bi et al, 2009;Wang et al, 2011;Hu et al, 2012). The PCR was conducted in 10 lL volume.…”

“…In addition, dinucleotide microsatellites are considered less efficient and more unreliable than tetranucleotides because of their minimal PCR stutter [ 25 ]. Moreover, the traditional methods of microsatellite isolation and characterization are quite involved, costly, and time-consuming [ 26 ]. With the publication of a number of genomes, we can obtain sufficient numbers of different types of useful microsatellite loci more efficiently [ 27 ].…”
