Isolation and Characterization of Three Antihypertension Peptides from the Mycelia of Ganoderma Lucidum (Agaricomycetes)

Abstract: Ganoderma lucidum (G. lucidum) has been widely used in Asia to treat hypertension, but the active substances responsible for its antihypertensive effects remain unclear. Using the well-established angiotensin I-converting enzyme (ACE) as a target, we identified three ACE inhibitory peptides (ACEIPs), Gln-Leu-Val-Pro (QLVP), Gln-Asp-Val-Leu (QDVL), and Gln-Leu-Asp-Leu (QLDL), which account for the antihypertensive activity of G. lucidum. Notably, QLVP worked in a mixed-type manner against ACE with an IC50 value… Show more

“…QLVP can inhibit ACE by its interaction with Gln242 and Lys472 of ACE. QLVP also enhanced Angiotensin 1-mediated phosphorylation of eNOS and reduced mRNA and protein expression of the vasoconstrictor peptide endothelin-1 in HUVECs (Wu et al, 2019).…”

“…Their computational protocol helped to explain that camel hemorphin had a higher binding affinity for ACE when compared to the mammalian one. In other work, Wu et al [27] identified three ACE inhibitory peptides, QLVP, QDVL, and QLDL, which account for the antihypertensive activity of Ganoderma lucidum, a polypore mushroom widely used in Asia to treat hypertension. They used docking and MD simulations to study the mode of action of the peptides.…”

“…The bioactive compounds are synthesized and secreted into the culture medium by the fermenting organisms during the process. SmF has been employed to ferment many food sources to obtain BPs, including cheese whey, mushroom, tomato pomace, and feather keratin (Mazorra-Manzano et al, 2020;Mechmeche et al, 2019;Moayedi et al, 2016Moayedi et al, , 2018Worsztynowicz et al, 2020;Wu et al, 2019). Several critical factors, including optimal operating conditions, substrates, and microorganisms used, should be considered during food fermentation for the production of BPs (Lasrado & Rai, 2018).…”
