Isolation and cross-species characterization of polymorphic microsatellites for the orchid bee Eulaema meriana (Hymenoptera: Apidae: Euglossini)

Abstract: We describe and characterize eight polymorphic microsatellite loci for the orchid bee species Eulaema meriana, an abundant species and important pollinator in wet lowland forests in tropical America. We also tested the cross-species amplification of these microsatellite loci in seven other species of the genus Eulaema. For E. meriana, number of alleles per locus ranged from four to nine and expected heterozygosity ranged from 0.377 to 0.854. Seven out of the eight loci described amplified in all seven other Eu… Show more

“…All mitochondrial and nuclear coding genes were sequenced in both directions, while the ASCN loci were sequenced only in the forward strand using a 24‐bp tag added to the forward primers (López‐Uribe et al . ). Fragments were sequenced using the Big Dye terminator kit (version 3.1) (Applied Biosystems) on an ABI 3730 capillary sequencer.…”

“…One of the reasons for this is a general lack of genetic markers for non-model organisms. Although the microsatellite loci of stingless bees are well studied (e.g., Francini et al, 2010;Francisco et al, 2011;Pereira et al, 2011) and there is a lot of literature on microsatellites in bumblebees (Estoup et al, 1995Funk et al, 2006, Stolle et al, 2009, microsatellites have only been developed for 10 species of solitary bees (Paxton et al, 1996(Paxton et al, , 2003Mohra et al, 2000;Kukuk et al, 2002;Azuma et al, 2005;Zayed, 2006;Souza et al, 2007;Soro & Paxton, 2009;Lopez-Uribe, 2011) and as a consequence there are few studies on the population structure and other biological aspects of these bees.…”
