Isolation and culture of murine bone marrow-derived macrophages for nitric oxide and redox biology

Abstract: Macrophages are mononuclear phagocytes derived from haematopoietic progenitors that are widely distributed throughout the body. These cells participate in both innate and adaptive immune responses and lie central to the processes of inflammation, development, and homeostasis. Macrophage physiology varies depending on the environment in which they reside and they exhibit rapid functional adaption in response to external stimuli. To study macrophages in vitro , cells are typically cultured… Show more

“…Emerging evidence suggests that Mϕ are involved in fibrotic responses. The Mϕ are a heterogeneous population with high plasticity, and can rapidly exhibit functional adaption in response to novel stimuli 32 . The expression of iNOS in Mϕ was increased by low molecular weight HA (LMW‐HA, 15–40 kDa) (Fig.…”

“…On day 7, the media were changed to serum-free Opti-MEM + 0.2% bovine serum albumin (BSA), to avoid the possibility that eicosanoids and other factors variably present in FBS might confound the results. 35 , 36 PMs were pretreated with vehicle or unlabeled apMPRO cells; and after 24 hours, labeled apMPRO cells were added to all cultures. The numbers of labeled apMPRO cells per PMs were determined as measures of 1° and 2° efferocytosis capacity.…”

“…Macrophages were derived from bone marrow following a published protocol [ 92 ]. Briefly, mouse bone marrow was flushed from the femurs and tibias of 8-week-old mice, plated in petri dishes in modified Dulbecco’s Modified Eagle Medium (DMEM).…”
