Antibiotic resistance continues to be an emerging threat both in clinical and environmental settings. Among the many causes, the impact of postchlorinated human wastewater on antibiotic resistance has not been well studied. Our study compared antibiotic susceptibility among Aeromonas spp. in postchlorinated effluents to that of the recipient riverine populations for three consecutive years against 12 antibiotics. Aeromonas veronii and Aeromonas hydrophila predominated among both aquatic environments, although greater species diversity was evident in treated wastewater. Overall, treated wastewater contained a higher prevalence of nalidixic acid-, trimethoprim-sulfamethoxazole (SXT)-, and tetracycline-resistant isolates, as well as multidrug-resistant (MDR) isolates compared to upstream surface water. After selecting for tetracycline-resistant strains, 34.8% of wastewater isolates compared to 8.3% of surface water isolates were multidrug resistant, with nalidixic acid, streptomycin, and SXT being the most common. Among tetracycline-resistant isolates, efflux pump genes tetE and tetA were the most prevalent, though stronger resistance correlated with tetA. Over 50% of river and treated wastewater isolates exhibited cytotoxicity that was significantly correlated with serine protease activity, suggesting many MDR strains from effluent have the potential to be pathogenic. These findings highlight that conventionally treated wastewater remains a reservoir of resistant, potentially pathogenic bacterial populations being introduced into aquatic systems that could pose a threat to both the environment and public health. IMPORTANCE Aeromonads are Gram-negative, asporogenous rod-shaped bacteria that are autochthonous in fresh and brackish waters. Their pathogenic nature in poikilotherms and mammals, including humans, pose serious environmental and public health concerns especially with rising levels of antibiotic resistance. Wastewater treatment facilities serve as major reservoirs for the dissemination of antibiotic resistance genes (ARGs) and resistant bacterial populations and are, thus, a potential major contributor to resistant populations in aquatic ecosystems. However, few longitudinal studies exist analyzing resistance among human wastewater effluents and their recipient aquatic environments. In this study, considering their ubiquitous nature in aquatic environments, we used Aeromonas spp. as bacterial indicators of environmental antimicrobial resistance, comparing it to that in postchlorinated wastewater effluents over 3 years. Furthermore, we assessed the potential of these resistant populations to be pathogenic, thus elaborating on their potential public health threat.

Controlling biofilm forming, pathogenic bacteria is an emerging challenge in the food industry, and the present study aims to test the efficacy of existing food preservation methods against these bacteria. We isolated 10 different biofilm forming pathogenic bacteria viz. Klebsiella quasipneumoniae, Bacillus cereus, Bacillus amyloliquefaciens, Bacillus anthracis, and Myroides odoratimimus from spoiled milk, biscuits, and cakes. K. quasipneumonia and B. amyloliquifaciens formed strong biofilm (OD 550 > 0.85), whereas B. cereus, B. anthracis, and M. odoratimimus made moderate biofilm (OD 5505 > 0.6) in 72 hr. The isolates, B. amyloliquifaciencs, B. anthracis, and M. odoratimimus were found positive for protease activity. The isolates showed significant tolerance to preservation conditions: pH (4-10), salt (up to 5%), temperature (up to 508C) and sodium benzoate (up to 0.1%).The isolates also exhibited high antibiotic resistance, maximum by M. odoratimimus (MIC 256 mg/ ml) against chloramphenicol which can have serious implications especially in tackling food borne illness caused by the biofilm forming food pathogens. Practical applicationsThe present study discloses the presence of biofilm forming pathogenic bacterial strains in common food stuff and their tolerance of preservation conditions and resistance to antibiotics. The occurrence of pathogens tolerant to common preservation conditions and their resistance to antibiotic should be addressed very cautiously. The significant findings of this study warrant the need for more stringent preservation methods to control food pathogens, especially the biofilm forming type. Moreover, findings of the present study will help to design novel preservation techniques such as incorporating antibiofilm coatings in food processing units and packaging materials, which will ensure food safety and public health.

Tetracycline Gene Transfer in Staphylococcus xylosus in situ During Sausage Fermentation

The presence of determinants of resistance to antibiotics can constitute a possible safety hazard in coagulase-negative staphylococci (CNS), which are widely present in food of animal origin. Among CNS, S. xylosus is a species frequently isolated from fermented meat products. Resistance to tetracycline was found to be one of the most distributed resistances occurring in S. xylosus strains isolated from fermented sausages. We evaluated the transfer of tetracycline resistance in vitro and in situ between S. xylosus strains. We selected three strains isolated from dry fermented sausages, resistant to tetracycline but not to minocycline, their resistance occurring by a mechanism of active efflux encoded by the tetK gene. Only one strain was able to transfer its tetracycline resistance to a recipient strain initially susceptible and plasmid-free using a filter mating procedure. Transfer of tetracycline resistance was observed at very low frequencies of 3.4 × 10−9 per recipient. To further investigate the transferability of this tetracycline resistance, the donor and recipient strains were tested in pilot-scale fermented sausage production. This transfer was possible but at a low rate, 1.4 × 10−7, and only under conditions of a high inoculation level of 108 CFU/g of meat. The tetK gene is located on a small mobilizable plasmid close to Staphylococcus aureus pT181 plasmid. In conclusion, the transfer of tetracycline resistance between strains of S. xylosus is possible, but at a really low frequency in vitro and in situ in fermented sausages. Even if this represents a very moderate risk, it should be taken into account as required by the European approach of Qualified Presumption of Safety (QPS) and AFSSA safety recommendations, advising that strains used as starter cultures should not carry any transferable antibiotic resistance.
