Isolation, Characterization and Electron Microscopy Analysis of A Hemidiscoidal Phycobilisome Type from the Cyanobacterium Anabaena sp. PCC 7120

Abstract: In this work we present the characterization of a hemidiscoidal phycobilisome type of the heterocystforming cyanobacterium Anabuena sp. PCC 7120. The phycobilisome of this organism contains allophycocyanin, phycocyanin and phycoerythrocyanin, similar to the closely related thermophilic cyanobacterium Mastigocladus laminosus. Intact phycobilisomes exhibit an absorption maximum at 619 nm and two fluorescence maxima at 664 nm and 680 nm, corroborating the presence of a complete energy pathway along the antenna. U… Show more

“…The two half core cylinders C1 and C2 are unique. In contrast to half core cylinders arranged in a side-by-side manner with other core cylinders in early models [5], C1 and C2 are arranged perpendicularly to the other core cylinders ( Figure 1A). In addition, these half core cylinders have connections to all other PBS components (core cylinders A1, A2, and B, and all peripheral rods), suggesting that they are important in the overall organization of this PBS.…”

“…Four morphological types of PBS 1 are known: hemidiscoidal 7 , hemiellipsoidal 8 , block-type 9 and bundle-type 10 . The hemidiscoidal PBS contains a central core surrounded by peripheral rods 1, 11,12 . The chromophores in hemidiscoidal PBS are arranged in such a way that a photon absorbed by a chromophore in the peripheral rods is rapidly funnelled to chromophores in the core 13 and eventually to the terminal emitters (the core-membrane linker protein (L CM ) [14][15][16] or allophycocyanin D (ApcD) 17,18 ).…”

“…The authors hypothesized that this form of PC was a connector between rods and the core, but this assertion was based only on the fact that this form of PC copurifies with some AP, and stoichiometric competence for this assertion was not demonstrated. If this type of "connector" were to be required to attach rods onto cores, then the extensive electron microscopic analyses performed on isolated PBS would probably have revealed this extra rod segment, and demethyl-PC should have accounted for a major fraction of the total PC (41)(42)(43)(44)(45)(46)(47)(48)(49). We have also observed small amounts (ϳ1-3%) of unmethylated PC subunits in Synechococcus sp.…”
