Isolation of a human prostate carcinoma cell line (DU 145)

Abstract: A long-term tissue culture cell line has been derived from a human prostate adenocarcinoma metastatic to the brain. The cell line, DU 145, has been passaged 90 times in vitro over a period of 2 years. The cells are epithelial, grow in isolated islands on plastic Petri dishes, and form colonies in soft agar suspension culture. Karyotypic analysis demonstrates an aneuploid human karyotype with a modal chromosome number of 64. Distinctive marker chromosomes (a translocation Y chromosome, metacentric minute chromo… Show more

“…The cell line DU-145 was originally derived from a brain metastasis of a human prostate adenocarcinoma (Stone et al, 1978); it retains the androgen independence of the original tumour and does not express a functional androgen receptor . This cell line possesses both LHRH and EGF receptors and produces EGFR ligands TGF-a and EGF (Xie et al, 1995;Jungwirth et al, 1997a).…”

“…The LNCaP and DU145 cell lines were originally isolated from metastatic lesions of patients suffering from prostate cancer (Stone et al, 1978;Horoszewicz et al, 1983) and purchased from DSMZ (Braunschweig, Germany) as American Type Culture Collection (ATCC) cell lines. HeLa cells were derived from a cervical carcinoma (Scherer et al, 1953) obtained from Dr Aubee.…”

“…38 Two androgennonresponsive cell lines were used: PC3 cells were derived from a human prostatic adenocarcinoma metastatic to the bone, 39 and DU-145 prostate carcinoma cells were derived from a human prostate adenocarcinoma metastatic to the brain. 40 LNCaP, DU-145, and PC3 cells were grown in DMEM supplemented with 30 mM 4-(2-hydroxyethyl)-1-piperazineethanesulfonic acid (HEPES), 10% FBS, 60 units/mL penicillin, and 60 units/mL streptomycin. The MDA-PCa-2b cells were cultured in Kaighn-modified Ham F12 medium with 2 mM L-glutamine that was modified by the ATCC to contain 1.5 g/L sodium bicarbonate.…”
