Isolation of Bluetongue Virus Serotype 1 from Culicoides vector Captured in Livestock Farms and Sequence Analysis of the Viral Genome Segment-2

Abstract: Bluetongue virus serotype-1 (BTV-1) was isolated from Culicoides oxystoma vectors captured on livestock farms in two places of Gujarat, India. The viruses were isolated on BHK-21 cells, which produced characteristic BTV-related cytopathic effects between 24 and 48 h post-infection. Virus antigen was demonstrated in infected cells at different passage by a BTV-specific sandwich ELISA. Further, polyacrylamide gel electrophoresis and silver staining of viral genomic RNA revealed ten double-stranded RNA segments c… Show more

“…Twenty-six different serotypes of BTV are recognized worldwide and distinct topotypes defined by closely related sequences of each genome segment have been proposed [19]. In India, 23 serotypes of BTV have been reported, identified either by serology or virus isolation, and eight serotypes (BTV-1, -2, -3, -9, -10, -16, -21, and -23) have been isolated from different regions during the last decade [4,5,8,12,17,22,26,28,30,33,36,37].BT is endemic in India. Multiple serotypes of BTV commonly circulate in the same geographical area, and can be isolated from the same flock or even from the same animals.…”

“…Other workers have reported the presence of BTV in various species of Culicoides from different countries [4,[26][27][28]. Earlier, BTV was isolated from pooled sample of Culicoides midges, collected from the western part of India [29,30].…”

“…Culicoides peregrinus has been reported as a potential vector of BTV in Yunnan Province in China, and in Australasia (Mellor et al ., ). The possibility that C. oxystoma plays a role as a vector of BTV was reaffirmed with the isolation of BTV serotype‐1 in the state of Gujarat in India (Dadawala et al ., ).…”
