Isolation of dopamine D2 receptor (D2R) promoters in Mugil cephalus

Abstract: This paper reports the isolation of two putative D(2)R promoters from grey mullet, one 5'flanking and the other an intronic sequence immediately upstream of the first coding exon. Promoter activity of the intronic sequence was confirmed in vitro through functional analysis using luciferase as reporter gene. The functional characteristics of the region flanking the 5'UTR is currently under investigation.

“…Taken together, our results suggest that the effect of estradiol on drd2 is part of the negative steroid feedback mechanism at vitellogenic stages. The recent finding that the promoter of mullet drd2 possesses a functional estradiol-responsive element [61] further corroborates our hypothesis.…”

“…51 In contrast, dopaminergic signaling via Drd2a was reported in Tilapia zillii, where an increase of drd2a mRNA paralleled an increase in plasma E2 levels, 47,48,52 suggesting the presence of ER-responsive elements (ERE) in the promoter of drd2a. 53 In the present study, we observed a decrease in drd2a mRNA with a corresponding increase in plasma E2 for high PFAS and low PAH/low PFAS treatment. Other combined exposures (high PAH/low PFAS, low PAH/high PFAS, and high PAH/high PFAS) did not produce any change in the expression of brain drd2a transcripts.…”

“…mykiss), binding of dopamine to Drd2a inhibited the production of GnRH-stimulated gonadotropin release from the pituitary. Elsewhere in goldfish (Carassius auratus), the release of GnRH-activated LH was blocked through Drd2a activation . “…In O. mykiss , the E 2 receptor is localized in three principal neuroendocrine territories: the ventral telencephalon, the anterior preoptic region and the mediobasal hypothalamus (Anglade et al , 1994), and the oestrogen receptor protein has been demonstrated to co‐localize with TH in a subpopulation of preoptic neurones known to produce DA and innervate the pituitary (Linard et al , 1995, 1996). Moreover, the promoter of the M. cephalus D2‐R has been recently shown to possess a functional E 2 ‐responsive element (Nocillado et al , 2006; Nocillado & Elizur, 2008). Therefore, sex steroids may exert direct actions on D2‐R gene transcription in teleosts.…”
