Isolation of single osteons and osteon lamellae

Abstract: Single human osteons were isolated by propagating fractures along their natural boundaries. Furthermore, decalcified osteons were mechanically manipulated to expose their interlamellar interfaces, making possible the study of collagen fibers by means of scanning electron microscopy (SEM). Isolation of single osteons of a wide range of lengths, as described in this paper, makes possible for the first time the study of their mechanical properties in all possible modes. SEM studies of exposed lamellar interfaces … Show more

“…Demineralized Samples Demineralisation of bone samples has been employed to permit interlamellar separation, thus making it easier to determine the preferred collagen bundle orientation in successive bone lamellae (Frasca et al, 1976). Archival demineralized bone histology material may also be investigated (Fig.…”

“…Part of the complexity arises from the size of the osteons and the inherent difficulty to their isolation. The methodology proposed in this paper seems to be less time consuming and as effective as the previous attempts by Ascenzi et al, (1959), (1967), (1985), (1994), Frasca et al, (1976), and Dong et al, (2005). Another component of the problem stems from the complexity of the analytical solution which requires the determination of both the matrix material properties and the drained elastic constants.…”

“…Ascenzi and coworkers (Ascenzi and Fabri 1959; Ascenzi and Bonucci 1967 Ascenzi et al, 1985; Ascenzi et al, 1994) developed a method for extraction of osteons from a thin cross section of bone using a sharp steel needle eccentrically inserted in a dentist’s drill or using a custom made micro lathe. Osteons have also been isolated by propagating a fracture through the natural boundary of the osteon (Frasca et al, 1976) and using a precision and computer controlled osteon pushout micro-testing system (Dong et al, 2005). …”

“…Frasca et al . separated and imaged lamellae in scanning electron microscopy, however, they did not make a distinction between primary and secondary osteons, nor image sufficient length to determine long-range three-dimensional lamellar shape [ 69 ]. Mohsin et al .…”
