Isolation of the Pharmacologically Active Saponin Ginsenoside Rb1 from Ginseng by Immunoaffinity Column Chromatography

Abstract: Immunoaffinity column chromatography using an anti-ginsenoside Rb1 monoclonal antibody has made possible a single-step separation of ginsenoside Rb1 from a crude extract of ginseng roots (Panax ginseng). The combination of immunoaffinity column chromatography and an enzyme-linked immunosorbent assay (ELISA) was also investigated.

“…True ginsengs are plants in the genus Panax from which Asian ginseng (Panax ginseng) and American ginseng (Panax quinquefolium) have received the most interest for phytomedicinal use. However, Eleutherococcus senticosis, a completely different plant not even in the genus Panax, is sometimes referred to as Russian or Siberian "ginseng" (Briksin 2000; Fukuda et al, 2000;Park et al, 2002;Ruan et al, 2010). There are two types of preparations from ginseng: white ginseng prepared by drying after peelling off and red ginseng prepared by steaming and drying (Shibata, 2001 (Park et al, 2002).…”

“…Furthermore, we have been establishing several affinity column with MAb against a kind of terpenoid, forskolin (Yanagihara et al, 1996), solasodine glycosides (Putalun et al, 1999), ginsenosides Rb1 (Fukuda et al, 2000c), and GC (Xu et al, 2007). In this chapter, we introduce the new strategy to isolate and concentrate the target bioactive compounds from the crude extract by immunoaffinity column using individual MAb.…”

“…Lane 1 and 2 indicate the standard of ginsenosides (G-Rd, G-Rc, G-Rb1, G-Rg1, and G-Re). Lane A, B, and C were the crude extract, the washing fraction, and the eluted fraction, respectively (Fukuda et al, 2000c) …”

“…In fact, we recently developed a new immunostaining method, known as Eastern blotting, against several glycosides (Tanaka et al 1997;Fukuda et al 2000c;Shan et al 2001). More recently, we reported the highly sensitive detection of benzopyrene using a surface plasmon resonance immunosensor (Miura et al 2002).…”
