Isolation of Two Novel Genes, Down‐regulated in Gastric Cancer

Abstract: Using a differential display technique, we identified two genes that are down-regulated in human gastric cancer tissue as compared to normal gastric mucosa. The down-regulated expression of these genes in gastric cancer tissue was confirmed by northern blotting analysis and RT-PCR. One, CA11, was a novel gene expressed predominantly in the stomach and was depleted in all of the gastric cancer cell lines examined. The other gene, GC36, was homologous to the digestive tract-specific calpain gene, nCL-4. The expr… Show more

“…These were used to create a cDNA probe for Northern blotting which confirmed the SAGE results. Simultaneously, the gene was identified by differential display by a Japanese group who designated it CA11 and reported the same expression pattern (Yoshikawa et al, 2000). CA11 is one of the most common transcripts in normal stomach, which suggests that it is functionally important, and demonstrates the power of mRNA profiling in the identification of novel genes.…”

“…Changes in the global pattern of gene transcription may be equally significant, and technological advances now permit their investigation (Carulli et al, 1998). Gastric carcinoma has been studied using differential display, Clontech cDNA membrane arrays and cDNA microarrays (Ebert et al, 2000;Jung et al, 2000;Yoshikawa et al, 2000;ElRifai et al, 2001;Wang et al, 2001;Hippo et al, 2002;Mori et al, 2002). However, we chose to use serial analysis of gene expression (SAGE), which produces comprehensive, quantitative and reproducible gene expression profiles (Velculescu et al, 2000).…”

“…The gene encoding this calpain was isolated as a recessive oncogene by using the antisense RNA strategy with NIH3T3 cells as a search host (Liu et al, 2000). Furthermore, it was reported that this gene is markedly down-regulated in di erentiated and poorly di erentiated types of gastric cancers (Yoshikawa et al, 2000).…”

“…The calpain family is divided into two groups, typical and atypical. The first group (calpains 1,2,3,8,9,11,12,14), are known as typical calpains because they are comprised of four domains including the Ca 2+ binding domain (domain IV). The second group (calpains 5,6,7,10,13,15) are known as atypical calpains because they lack the Ca 2+ binding domain (domain IV) [1].…”
