Janus kinase 3 regulates renal 25-hydroxyvitamin D 1α-hydroxylase expression, calcitriol formation, and phosphate metabolism

Abstract: Calcitriol, a powerful regulator of phosphate metabolism and immune response, is generated by 25-hydroxyvitamin D 1α-hydroxylase in the kidney and macrophages. Renal 1α-hydroxylase expression is suppressed by Klotho and FGF23, the expression of which is stimulated by calcitriol. Interferon γ (INFγ) regulates 1α-hydroxylase expression in macrophages through transcription factor interferon regulatory factor-1. INFγ-signaling includes Janus kinase 3 (JAK3) but a role of JAK3 in the regulation of 1α-hydroxylase ex… Show more

“…Moreover, the difference between B lymphocytes from jak3 -/-mice and jak3 +/+ mice may be affected by an influence of JAK3 deficiency on the abundance of inflammatory cells and the release of inflammatory mediators modifying BK channel activity. Along those lines enhanced serum calcitriol and FGF23 levels have been observed in jak3 -/-mice [57]: Moreover, JAK3-deficient mice are volume depleted [48].…”

“…In JAK3 deficient mice spleen, peripheral lymph nodes, and thymus are hypoplastic, and intestinal Peyer's patches completely lacking [18]. Jak3-deficiency leads to increased formation of 1,25(OH) 2 D 3 in the kidney [19] and to enhanced release of IL-12 [20], which is produced by several cell types including dendritic cells [21].…”

“…The pleotropic effects of JAK3 include regulation of transport across the cell membrane [43,45,49,50,51,52,53,54] including the excitatory amino acid transporters [53] and the Na + /K + ATPase [49]. …”
