Kaposi's Sarcoma–Associated Herpesvirus-Like DNA Sequences in AIDS-Related Body-Cavity–Based Lymphomas

Abstract: The recently discovered KSHV DNA sequences occur in an unusual subgroup of AIDS-related B-cell lymphomas, but not in any other lymphoid neoplasm studied thus far. Our finding strongly suggests that a novel herpesvirus has a pathogenic role in AIDS-related body-cavity-based lymphomas.

“…4 While a controversial increased incidence of multiple myeloma and other hematopoietic neoplasms in Classic Kaposi Sarcoma has been extensively reported in the literature, 14,16,18,[21][22][23][24][25][26][27][28][29] the suggested link between HHV-8 and multiple myeloma 30 supports an increase, such as noted in our study, in multiple myeloma in Kaposi Sarcoma patients. An increased incidence of other hematopoietic neoplasms was also observed in our cohort.…”

“…Primary effusion lymphoma (PEL) was first identified as a subset of body-cavity-based lymphomas, which subsequently was called PELs [47]. PELs are unique as they were found to contain KSHV DNA are most frequently found in men and in AIDS patient.…”

“…HHV-8 is present in all Kaposi's sarcoma lesions and is also associated with the rare AIDS-related body cavity-based B-cell lymphoma, non-AIDS-associated body cavity-based B-cell lymphomas, and multicentric Castleman's disease [Cesarman et al, 1995Boshoff et al, 1995;Huang et al, 1995;Soulier et al, 1995]. HHV-8 was detected in 8% of infants<15 months of age in Zambia, where a linear increase in seroprevalence with age was noticed that reflects the endemic nature of HHV-8 infection (47% in 14-19-year-olds to 71% in persons aged≥50 years) [Kasolo et al, 1997;Olsen et al, 1998].…”
