Key enzymes catalyzing glycerol to 1,3-propanediol

Abstract: Biodiesel can replace petroleum diesel as it is produced from animal fats and vegetable oils, and it produces about 10 % (w/w) glycerol, which is a promising new industrial microbial carbon, as a major by-product. One of the most potential applications of glycerol is its biotransformation to high value chemicals such as 1,3-propanediol (1,3-PD), dihydroxyacetone (DHA), succinic acid, etc., through microbial fermentation. Glycerol dehydratase, 1,3-propanediol dehydrogenase (1,3-propanediol-oxydoreductase), and … Show more

“…In the glycerol anaerobic pathway, glycerol uptake facilitator protein, glycerol dehydrogenase, glycerol dehydratases with corresponding reactivation factors, and 1,3-propanediol oxidoreductase are the key enzymes for 1,3-PDO formation (31). The significant upregulation of these genes might guarantee the activities of corresponding enzymes for the 1,3-PDO production in crude glycerol in response to stresses, for which the upregulation of some was in agreement with the similar levels of corresponding enzyme activities detected in the crude glycerol cultures.…”

“…Glycerol dehydratases encoded by three dhaB genes are regarded as key rate-limiting enzymes for catalyzing the dehydration of glycerol to 3-hydroxypropionaldehyde (3-HPA) in the reductive pathway (31). The transcript levels of the three dhaB genes in K. pneumoniae 2e were all significantly upregulated in crude glycerol relative to that in pure glycerol, whereas the genes encoding propanediol dehydratase, which is regarded as the functional substitution of glycerol dehydratase, were all downregulated.…”

“…[32] Until afew years ago, there were only chemical processes for the industrialp roduction of 1,3-PDO, starting from oil-based 1,2-EDOa nd acrolein. “…As the name implies, the overexpression of genes is an abnormally and excessively high level of gene expression which produces a pronounced gene-related phenotype (46). The salient feature of overexpression is that the process can be carried out not only in the original cells where it exists (homologous host), but also in another expressing system (heterologous host).…”

“…When the byproduct pathways were deleted, the intracellular redox balance (NADH/NAD + ) was extremely high in these mutants, which has already been confirmed by some researchers. Therefore, these groups investigated further how to overexpress genes dhaB and dhaT to use these saved NADH (46,48).…”
