Key role of 5-HT 3 receptors in the nucleus tractus solitarii in cardiovagal stress reactivity

Abstract: Serotonin plays a modulatory role in central control of the autonomic nervous system (ANS). The nucleus tractus solitarii (NTS) in the medulla is an area of viscerosomatic integration innervated by both central and peripheral serotonergic fibers. Influences from different origins therefore trigger the release of serotonin into the NTS and exert multiple influences on the ANS. This major influence on the ANS is also mediated by activation of several receptors in the NTS. In particular, the NTS is the central zo… Show more

“…Vagally-mediated RSA can be modulated by superior central influences through their actions on vagal motor neurons. For example, direct and indirect influences of central structures involved in stress, like the dorsomedial nucleus of the hypothalamus (DiMicco et al, 2002;Sévoz-Couche et al, 2013, 2003Sévoz-Couche and Brouillard, 2017), decrease the baroreflex gain and reduce the tonic baroreceptor drive on vagal motor neurons. This results in the diminution of the basal level of vagal activity and therefore the decrease of RSA (Brouillard et al, 2020(Brouillard et al, , 2019Sévoz-Couche et al, 2013, 2003.…”

“…Correlation between serotonin and the ANS, both of which have similar regulatory functions, has been reported previously 15 19 . Structurally, serotonergic neurons originating from the raphe nucleus in the midbrain project onto the nucleus tractus solitarius, which is responsible for cardiovascular reflexes and ANS function 15 , 16 . Clinically, the tryptophan depletion test, which decreases the serotonin level, may lead to lower HRV and more severe depressive symptoms 17 .…”

“…Glutamatergic transmission in the NTS is modulated by a complex variety of other transmitters (see Sevoz-Couche & Brouillard, 2017), with 5-hydroxytryptamine (serotonin, 5-HT) considered to play a key role in this important cardiovascular sensory nucleus (Ramage and Villalón, 2008; Hosford and Ramage, 2019). The transmitters and receptors involved in neuron-neuron communication necessary for reflex integration have been well studied.…”
