Kibdelins (AAD-609), novel glycopeptide antibiotics. II Isolation, purification and structure.

Abstract: A new glycopeptide antibiotic complex was isolated from the fermentation culture of Kibdelosporangium aridum subsp. largum (SK&F AAD-609) by affinity chromatography on a D-alanyl-D-alanine agarose column. The major components of the complex were resolved by preparative reversed-phase HPLC. Mild acid hydrolysis showed that the new antibiotics have the same mannosyl aglycon (2) as the aridicins.FAB mass spectrometry, isoelectric focusing, potentiometric titration and carbohydrate and fatty acid analyses were use… Show more

“…The acyl groups were nundecanoic acid in PA and A and isododecanoic acid in PB and B. Among the glycopeptides, teicoplanin, aridicins, and kibdellins also contain fatty acid moieties (3,12,27). Another similarity between A40926 and the aridicins was the presence of two carboxylic acid functions which gave these molecules a net negative charge at neutral pH.…”

“…PA and PB, described here for the first time, predominated in the broth; during purification, these two compounds were largely converted to A and B, whose structures have been described recently (31a). One feature of all four factors, which relates them to teicoplanin and some other glycopeptides (aridicins and kibdellins) (3,27,12) while distinguishing them from vancomycin, ristocetin, and others, is the presence of a fatty acid moiety attached to one of the sugars. In terms of biological activity, the most striking difference between the components of the A40926 complex and other glycopeptide antibiotics is their activity against Neisseria gonorrhoeae.…”

“…Several glycopeptides containing a long-chain aliphatic acyl residue on one of the amino sugars have been reported (8,9,19,32,39). Some of these compounds were reported to be superior in antibacterial activity and pharmacokinetics to vancomycin (10).…”

“…Members of the genus Kibdelosporangium , like Kibdelosporangium aridum subsp. largum and Kibdelosporangium philippinense , were found to produce antibiotics such as aridicins A [9], B and C [10], and kibdelins A, B [11], C and D [4], respectively.…”
