Kinetics of Inhibition Effect of 4-hydroxy-3-methoxybenzoic acid on Mushroom Tyrosinase

Abstract: A study on the kinetics of inhibitory effect of 4-hydroxy-3-methoxybenzoic acid on the monophenol- and diphenol-oxidation activity of mushroom tyrosinase has been carried out using enzymological kinetic analysis method in a Na2HPO4-NaH2PO4 buffer solution (pH=6.8) at 30 °C. The results show that 4-hydroxy-3-methoxybenzoic acid efficiently can inhibit both monophenol- and diphenol-oxidation activity of mushroom tyrosinase under experiment conditions. Concentrations of 4-hydroxy-3-methoxybenzoic acid leading to … Show more

“…The determination of the total phenolic content (TPC) was based on the method described by Ainsworth et al [88] and Bridi et al [89], with some modifications. Briefly, the mixture of 10% (v/v) Folin-Ciocalteu reagent and leaves extract dissolved in ethanol was incubated for 5 min at 25 • C. After that, 350 mM sodium carbonate was added.…”

“…Standard solutions: Kojic acid, tropolone, isoferulic acid, resveratrol, and benzoic acid, all of which are described in the literature as tyrosinase inhibitors, were used as reference materials [1,[12][13][14]. Octinoxate [2] and dibutylphtalate were used as negative controls.…”

“…It is evident that tyrosinase is a key enzyme controlling the melanization process of skin, eye, inner ear and hair, as well as the enzymatic browning process in fruits and vegetables [3,4,5]. Meanwhile, the application of tyrosinase activators has attracted more and more attentions in the fields of cosmetic, food and pharmaceutical industry, primarily because of their high efficacy in mitigating hyperpigmentation [6].…”
