Klebsiella pneumoniae—A Useful Pathogenic Strain for Biotechnological Purposes: Diols Biosynthesis under Controlled and Uncontrolled pH Levels

Abstract: Despite being a well-known human pathogen, Klebsiella pneumoniae plays a significant role in the biotechnology field, being considered as a microbial cell factory in terms of valuable chemical biosynthesis. In this work, Klebsiella pneumoniae DSMZ 2026 was investigated for its potential to biosynthesize 1,3-propanediol (PDO) and 2,3-butanediol (BDO) during batch fermentation under controlled and uncontrolled pH levels. The bacterial strain was cultivated at a bioreactor level, and it was inoculated in 2 L of s… Show more

“…The organic acid separation was accomplished on a reversed-phase chromatographic column Acclaim OA (5 µm, 4 × 150 mm Dionex), washed with NaH2PO4 50 mM concentration (pH 2.8) solution for 10 min, at 20 °C, and with a 0.5 mL/min flow rate. Chromatogram measurement was performed at a λ = 210 nm wavelength [ 46 , 47 ]. Organic acid standard stock solution (Merck) was prepared by mixing the components (lactic, tartaric, malic, citric, succinic, fumaric, butyric acids) ( Supplementary Figure S1 ).…”

“…Among the tested polyols within this work, BDO is less studied as a plasticizer in PVOH-based biofilms, even though it is largely used as an intermediate compound in hard rubber product manufacturing, and also in the pharmaceutical industry as a mediator [70].…”
