Klotho Regulation of TRPC Channels Involving Human Breast Cancer Cell Migration

Abstract

Klotho is a mammalian aging‐suppression gene and function as a suppressor of cancer. Here, we investigated the molecular mechanism of Klotho regulation of TRPC channels involving human breast cancer cell migration. Serum‐induced activation of TRPC and cell migration was specifically blocked by TRPC channel inhibitor, SKF96365, but not by voltage‐gated Ca2+ channel inhibitor, nimodipine. Treatment of purified Klotho protein inhibited TRPC6 channel activity. Interestingly, serum treatment increased the surface expression of TRPC6 channel. Klotho suppressed cell surface abundance of TRPC6 channels, whereas did not affect gating and activation kinetics of the channel. In addition, treatment of PI3K inhibitor (Wortmannin) or Erk1/2 inhibitor (U0126) blocked the serum‐induced augmentation of TRPC6. Suppression of vesicular trafficking by treatment of brefeldin A or tetanus toxin significantly decreased TRPC6 activity and Klotho regulation.Taken together, our data suggest that Klotho decreases surface expression of TRPC6 probably by reducing serum growth factor‐induced secretory pathway of the channel.
