Knockout of Secretin Receptor Reduces Large Cholangiocyte Hyperplasia in Mice With Extrahepatic Cholestasis Induced by Bile Duct Ligation

Abstract: During bile duct ligation (BDL), the growth of large cholangiocytes is regulated by the cyclic adenosine monophosphate (cAMP)/extracellular signal-regulated kinase 1/2 (ERK1/ 2) pathway and is closely associated with increased secretin receptor (SR) expression. Although it has been suggested that SR modulates cholangiocyte growth, direct evidence for secretin-dependent proliferation is lacking. SR wild-type (WT) (SR) mice underwent sham surgery or BDL for 3 or 7 days. We evaluated SR expression, cholangiocyte … Show more

“…The cholangiocyte proliferative response in these animal models is associated with increased SCTR gene expression and elevated secretinstimulated cAMP levels. Furthermore, administration of secretin (2.5 nmol·kg body wt Ϫ1 ·day Ϫ1 ) intraperitoneally by osmotic minipumps for 1 wk stimulates the proliferation of large cholangiocytes, while the knockout of the SCTR inhibits their proliferation following bile duct ligation by ϳ50% (14).…”

“…Bile ducts are known to proliferate in response to ligation of the main duct as part of what is believed to be a compensatory response. Recently, this proliferative response in large cholangiocytes was shown to be absent in Sctr Ϫ/Ϫ mice (16). Whether this response is triggered by increased pressure or cholestasis is unclear.…”
